Position Summary
We seek a highly motivated student to join our Predictive Sciences group within the Oncogenesis Thematic Research Center for a 10-week summer internship program. A successful candidate will work with their mentor to interrogate the biological functions of oncogenic transcription factors Ikaros and Aiolos in Diffuse Large B Cell Lymphoma (DLBCL) by a retrospective analysis of multi-modal omics data.
DLBCL is an aggressive heterogeneous disease with poor prognosis associated with cell of origin or high-risk molecular features. Ikaros and Aiolos have been identified as key oncogenic drivers for DLBCL. Over the years, BMS has developed multiple cereblon (CRBN) modulating agents, from first-generation compounds like lenalidomide and pomalidomide to more recent golcadomide, to induce targeted degradation of Ikaros and Aiolos by hijacking the ubiquitin proteasome system. However, the specific regulatory roles and biological functions of Ikaros and Aiolos in DLBCL remain obscure.
Recent advances in CUT&RUN have enabled high-quality profiling of Ikaros and Aiolos' binding landscape. With the CUT&RUN capability being established within the Oncogenesis TRC and a large collection of genetic and epigenetic profiling we have accumulated over the years for a panel of DLBCL cell models (e.g. ChIP-seq, ATAC-seq, Micro-C/Hi-C, RNA-seq WGS, WGBS and genome-wide CRISPR screen), we expect the successful candidate to apply and/or develop effective data-driven methods to analyze, mine, and interpretate the data towards a better understanding of Ikaros and Aiolos' biological functions in DLBCL.
As a leading company in Targeted Protein Degradation (TPD), this intern will provide an exciting opportunity for the candidate to have an immersive experience of the development of protein degrader drugs. The candidate will also have opportunities to learn from experienced computational and experimental biologists and hone the skills in applying data-driven methods to tackle challenges in new drug development.
Key Responsibilities
  • Perform in-depth analysis of CUT&RUN data to map the cell-type-specific binding landscape of Ikaros and Aiolos across a panel of DLBCL models and generate hypotheses linked to phenotype (e.g. sensitivity to Ikaros and Aiolos' degradation).
  • Contextualize the hypothesized functions of Ikaros and Aiolos within the genetic and epigenetic landscapes of a panel of DLBCL cell models by data-driven methods and literature search.
  • Deliver and communicate analyses results to the team.

Qualifications & Experience
  • Currently enrolled in a Ph.D program in physical sciences, life sciences, bioinformatics, computer sciences, or a related field.
  • Strong interests in solving biological problems by applying quantitative skills.
  • Hands-on experience in genetic and/or epigenetic profiling techniques (e.g. CUT&RUN, ChIP-seq and WGBS) is required.
  • Experience in Python programming language (e.g. pandas, numpy) or R.
  • Familiarity with public omics data consortiums (e.g. TCGA, DepMap and ENCODE) is preferred but not required.
  • Familiarity with enhancer biology and higher-order chromatin structure is preferred but not required.
  • Must be able to work full-time for the program duration.
  • Excellent communication and presentation skills.
